Identification of Tomato yellow leaf curl virus-IR and associated Tomato leaf curl betasatellite infecting common bean (Phaseolus vulgaris) in Oman.

Abstract

In 2016, foliar yellow mosaic, severe curling, and crumpling symptoms, with an incidence of 20 to 35%, were observed on bean (P. vulgaris) plants in mixed crop (tomato, chili and watermelon) fields in the Al-Batina region of Oman together with a high-density population (>10 adults/leaf) of Bemisia tabaci (MEAMI). Five bean plants with prominent symptoms and two nonsymptomatic plants were collected from two farms ∼5 km apart. An initial analysis of crude sap extracted from symptomatic leaf samples under electron microscopy with negative staining showed the presence of twinned-icosahedral particles (18 to 20 nm) typical of geminiviruses. PCR assay and rolling-circle amplification were carried out. BLAST comparison of the sequence to sequences available in the GenBank nucleotide sequence database showed 98% nt identity to an isolate of the "Iran" strain of Tomato yellow leaf curl virus (TYLCV; FJ956703) previously identified infecting tomato in Oman. PCR amplification using universal betasatellite primers showed the presence of a betasatellite in all five plants. For one sample, the PCR product was cloned and sequenced. The amplicon contained 1 366 nt and showed an arrangement typical of betasatellites, with a single conserved gene (βC1) in the complementary-sense. Sequence comparisons showed the sequence to have 99% nt identity to an isolate of tomato leaf curl betasatellite (ToLCB) previously identified in Oman. This is thought to be the first record of TYLCV-Iran, in conjunction with ToLCB, infecting P. vulgaris in Oman.
